Protonix accumulates in the tubules of parietal cells of the stomach and transforms into its active form, which selectively interacts with the H+-K+-ATP-ase enzyme. By inhibiting this enzyme, Protonix prevents the transfer of hydrogen ions from the parietal cells into the stomach cavity, effectively blocking the final stage of hydrochloric acid secretion. It inhibits both basal and stimulated secretion of hydrochloric acid, regardless of the stimulus type (acetylcholine, histamine, or gastrin). Protonix is used to treat:
Take Protonix exactly as prescribed by your doctor. The typical dosage for stomach or duodenum ulcers and gastroesophageal reflux disease is 40 mg once daily, usually for four weeks. For combined treatment with antibiotics (amoxicillin and clarithromycin) to eradicate H. pylori, take Protonix 40 mg daily divided into two doses: one tablet before breakfast and one before supper. Swallow the tablet whole with a glass of water; do not crush, chew, or break it. If using Protonix oral granules, mix them only with applesauce or apple juice for easier swallowing. Do not use other liquids or soft foods.
Long-term use of Protonix may be necessary for some conditions. While this medication has been linked to cancer in animal studies, its effects on humans are unknown. Protonix may also cause a vitamin B-12 deficiency, with symptoms such as pale skin, weakness, tiredness, shortness of breath, and a fast heart rate. Consult your doctor if you are pregnant, planning to become pregnant, or breastfeeding, as there is no evidence on the drug's effects on unborn babies.
Protonix should not be used if you have:
Common side effects of Protonix include:
Allergic reactions such as hives, facial or tongue swelling, and rash may also occur.
Protonix may reduce the absorption of pH-dependent medications like ketoconazole, decreasing their effectiveness. It can also increase the absorption and blood concentration of digoxin (Lanoxin), potentially leading to toxicity. Inform your doctor about all medications you are taking to avoid interactions.
If you miss a dose, take it as soon as possible. If it is almost time for your next dose, skip the missed dose and resume your regular schedule. Do not double the dose to compensate.
An overdose of Protonix is not expected to be life-threatening. However, if you suspect an overdose, seek immediate medical assistance.
Store Protonix at room temperature (15-30°C or 59-86°F), away from light, moisture, children, and pets.
