Common Use
Clozaril is an atypical antipsychotic medication used to treat schizophrenia in patients who do not respond to other medicines. It helps reduce the risk of suicidal behavior by affecting certain chemicals in the brain.
Dosage and Directions
Take Clozaril exactly as prescribed by your doctor. It can be taken with or without food, but taking it with food may help reduce stomach irritation if upset occurs. Continue taking the medication even if you feel well, and do not stop suddenly without consulting your doctor.
Precautions
Inform your doctor about all medical conditions, especially if you:
- Are pregnant, planning to become pregnant, or breastfeeding.
- Have allergies, a history of blood clots, blood problems, high cholesterol, heart issues, or lung or breathing problems.
- Have a history of enlarged prostate, prolonged painful erections, liver or kidney problems, seizures (e.g., epilepsy), glaucoma, stomach or bowel issues, or dementia.
- Have poor health, diabetes, a family history of diabetes, or are overweight.
- Have decreased activity of enzymes that metabolize certain medicines.
Contraindications
Do not take Clozaril if you:
- Are allergic to it.
- Experience severe drowsiness, uncontrolled seizures (epilepsy), or loss of bowel muscle movement (paralytic ileus).
- Have bone marrow problems, other blood cell issues, or a history of blood problems caused by Clozaril.
- Are taking other medications that may cause blood problems.
Possible Side Effects
Common side effects include:
- Dry mouth, headache, dizziness, heartburn, increased sweating or saliva production, lightheadedness when standing, nausea, strange dreams, trouble sleeping, and weight gain.
Severe side effects may include:
- Severe allergic reactions (rash, hives, difficulty breathing, chest tightness, swelling of the mouth, face, lips, or tongue).
- Agitation, calf pain or tenderness, chest pain, confusion, dark urine, decreased coordination, fainting, fast or irregular heartbeat.
- Fever, chills, sore throat, increased hunger, thirst, or urination, infection, involuntary movements of the tongue, face, mouth, or jaw.
- Loss of appetite, numbness of an arm or leg, rapid breathing, restlessness, seizures, severe headache, dizziness, or vomiting.
- Severe or persistent nausea or constipation, severe stomach pain, shortness of breath, stiff muscles, sudden weight gain, swelling of the hands, ankles, or feet, tremor, trouble swallowing, uncontrolled muscle movements, unusual mental or mood changes, unusual tiredness or weakness, vision changes, vomiting, or yellowing of the skin or eyes.
Drug Interactions
Inform your doctor about all medications you are taking, especially:
- Benzodiazepines (lorazepam) or other medicines for mental or mood problems, as they may increase the risk of heart or breathing issues.
- Cimetidine, erythromycin, quinolone antibiotics (ciprofloxacin), risperidone, or SSRIs (e.g., fluoxetine), as they may increase the risk of Clozaril's side effects.
- Barbiturates (phenobarbital), hydantoins (phenytoin), nicotine, or rifampin, as they may decrease Clozaril's effectiveness.
- Antiarrhythmics (e.g., propafenone, quinidine), anticholinergics (benztropine), carbamazepine, debrisoquin, dextromethorphan, medicines for depression or high blood pressure, phenothiazines (chlorpromazine), or tricyclic antidepressants (amitriptyline), as Clozaril may increase the risk of their side effects.
Missed Dose
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and return to your regular schedule. Do not double the dose. If you miss taking Clozaril for more than 2 days, contact your doctor immediately for instructions.
Overdose
Symptoms of overdose include drowsiness, slow or difficult breathing, dizziness, excessive salivation, fainting, coma, or fast or irregular heartbeats. Seek medical attention immediately if an overdose is suspected.
Storage
Store Clozaril at room temperature, between 59 and 86 degrees F (15 and 30 degrees C). Keep it away from heat, moisture, and light. Do not store in the bathroom. Keep out of reach of children and pets.
